Urinary proteins are important for many application such as Biomarker discovery, differential expression of proteins in various diseases, and diagnosis. The ProteoSpin™ Urine Protein Concentration Maxi Kit is able to concentrate urine proteins several fold as well as remove salts, urea, and other urine contaminants. This kit has no molecular weight cut-off and therefore captures total urinary proteins and peptides of all sizes, making it ideal for biomarker discovery work or other diagnostic research work. This is an advantages over other commercial membrane devices and kits with molecular weight cutoffs that limit the size of the purified proteins. The kit employs spin column technology that offers convenience, speed, and ease-of-use, and therefore offers significant time savings over dialysis protocols.
The kit employs a proprietary matrix which acts as an ion exchanger and is able to concentrate proteins many fold as well as remove contaminants. Breifly, pH Binding Buffer is added to the protein sample and the sample is applied to Norgen's column (BIND). Under these conditions the proteins will bind to the column while salts and other contaminants are removed in the flowthrough. The bound proteins are then washed to remove any remaining impurities (WASH). Lastly, the purified urine proteins are eluted into a small volume of the provided Elution Buffer (ELUTE). Please see the procedure flowchart to the right Each spin column is able to concentrate and desalt up to 4 mg of urine proteins. The resulting high-quality protein sample is concentrated and free from the original sample salts, thus preparing the sample conveniently for downstream applications including SDS-PAGE, 2D SDS-PAGE, Whole protein mass spectrometry, MALDI-TOF, LC/MS, LC/MS/MS, Western blotting and Protein microarrays FEATURES AND BENEFITS

Figure 1. No Loss of Urine Proteins. Norgen’s Urine Protein Concentration Maxi Kit was used to concentrate a 20 mL urine sample according to the provided bind, wash and elute procedure. The urinary proteins present in the 20 mL urine sample were concentrated into 2 mL of the provided Elution Buffer. The urinary proteins were resolved on a 12% SDS-PAGE gel run at 200 V for 75 minutes, and were stained used Commassie Blue staining. In the gel above, Lane 1 contains 25 µL of input urine before concentration, Lane 2 contains 15 µL of the binding flowthrough, and Lane 3 contains 15 µL of the concentrated eluted protein. No urine proteins are lost during the binding step, as can be seen by examining the binding flowthrough. |
Figure 2. Concentration of Urine Proteins. Urine proteins are concentrated many-fold using the ProteoSpin™ Urine Protein Concentration Maxi Kit. The urinary proteins present in 20 mL of urine were concentrated into 2 mL of Elution Buffer using Norgen’s Urine Protein Concentration Maxi Kit according to the provided bind, wash and elute procedure. The urinary proteins were resolved on a 12% SDS-PAGE gel run at 200 V for 75 minutes, and the urinary proteins were stained used Commassie Blue staining. Lane 1 contains 20 µL of the urine sample that was used as the input, Lane 2 contains 5 µL of the elution, Lane 3 contains 10 µL of the elution, and Lane 4 contains 20 µL of the elution. By comparing Lanes 1 and 4, which contain equal volume amounts of input and output, it can clearly be seen that the urine proteins have been highly concentrated though the use of the kit. |
